Abstract:

Background: Intracytoplasmic sperm injection (ICSI) can be considered the solstice in vitro insemination technique because it has powerfully allowed the treatment of male factor infertility, but in this procedure, except visual morphological selection, there is no standardization for sperm selection. Recently, hypo osmotic swelling test (HOST) has been proposed to have the potential to select intact sperm. Therefore, the aim of this study was to evaluate the ability of this technique to select functional sperm in terms of apoptosis, morphology, nuclear and membrane integrity. Materials and Methods: Twenty semen samples were randomly collected from men attending the andrology unit. Semen samples were washed, exposed to hypotonic condition, then fixed and simultaneously assessed for abnormal morphology, DNA fragmentation and protamine deficiency using papanicoulau, TUNEL and CMA3 staining, in different HOST grades, respectively. For the assessment of membrane integrity or early apoptosis, the remaining semen samples were washed with Ca buffer and stained by Annexin V. Subsequently, Annexin stained samples were exposed to hypo osmotic conditions. Results: Healthy sperm were observed in HOST graded “d” and “c” while sperm with HOST grade “g” presented the highest degree of anomalies and frequency of apoptosis, morphology, nuclear and membrane integrity. Conclusion: Addition of HOST into the sperm selection procedure may provide a valuable tool for selection of functional sperm required for ICSI. Furthermore, selection of sperm with HOST grade “g” should be avoided during ICSI.

Is there an association between HOST grades and sperm quality?

BACKGROUND Intracytoplasmic sperm injection (ICSI) is the primary treatment for male infertility. However for this procedure, with the exception of visual morphological selection, there is no standardization for sperm selection. Recently, the hypo-osmotic swelling test (HOST) has been proposed to potentially select sperm with intact membranes. The aim of this study is to evaluate the ability of...

P-77: HOST Patterns and Sperm Quality

Background: Following the expansion of human infertility, different procedures have been utilized for separation of normal sperm during ICSI (Intra cytoplasmic sperm injection). Hypo-osmotic swelling test (HOST) is one of the tests which have been introduced for assessment of integrity of sperm membrane when confronted with hypo osmotic condition. This test may also reflect intracellular organe...

Is There any Association between Thyroid Autoimmunity and Breast Cancer?

Background: The relationship between thyroid autoimmunity and breast cancer is a challenging subject. We aimed to investigate this association in women with breast cancer. Methods: In this descriptive study, 41 women with newly diagnosed breast cancer before receiving any pharmacologic treatment and 38 healthy age-matched women were enrolled. Anti TPO Ab (anti-thyroid peroxidase antibodies), FT...

The association between leukocytes and sperm quality is concentration dependent

BACKGROUND To evaluate the association between leukocytes (polymorphonuclear granulocytes -PMNL) and semen parameters at different leukocyte concentrations. METHODS This was a retrospective clinical study at a university hospital andrology clinic. Semen samples from infertile men were analyzed for sperm morphology and motility according to seminal leukocytes (PMNL) concentration (category A: ...

Is there an association between ankylosing spondylitis

Ankylosing spondylitis (AS) is predominantly a disease of the spine and large joints but may be associated with extra-articular involvement. For example, cardiac, pulmonary and other systemic complications are well recognised.1 A variety of neurological syndromes have been described, including the cauda equina syndrome2 3 and, more recently, an association with multiple sclerosis (MS) has been ...
